Platelet Derived Growth Factor AA (PDGF-AA)

PDGFs are disulfide-linked dimers consisting of two 12.0-13.5kD polypeptide chains, designated PDGF-A and PDGF-B chains. The three naturally occurring PDGFs. (PDGF-AA, PDGF-BB and PDGF-AB) are potent mitogens for a variety of cell types including smooth muscle cells, connective tissue cells, bone and cartilage cells, and some blood cells. The PDGFs are stored in platelet alpha-granules and are released upon platelet activation. The PDGFs are involved in a number of biological processes, including hyperplasia, chemotaxis, embryonic neuron development, and respiratory tubule epithelial cell development. Two distinct signaling receptors used by PDGFs have been identified and named PDGFR-alpha and PDGFR-beta. PDGFR-alpha is high-affinity receptor for each of the three PDGF forms. On the other hand, PDGFR-beta interacts with only PDGF-BB and PDGF-AB. PDGF is released when platelets adhere to traumatized tissues. Connective tissue cells near the traumatized region respond by initiating the process of replication.The synthesis of PDGF can be induced by Interleukin-1, Interleukin-6, TNF-alpha, TGF-beta and EGF.\n\nApplications: \nSuitable for use in Neutralization, ELISA and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nNeutralization: 0.17-0.19ug/ml yields one-half maximal inhibition [ND50] of the biological activity (10ng/ml).\nELISA: > 0.5ug/ml detects 0.2-0.4ng/well by direct ELISA (using 100ul/well antibody solution).\nWestern Blot: 0.1-0.2ug/ml Detection limit is 1.5-3.0ng/lane, under either reducing or non-reducing conditions. \nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nLyophilized powder may be stored at 4 degrees C for short-term only. Reconstitute to nominal volume by adding sterile 40-50% glycerol and store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n\nCountry of Origin: \nUSA
