SHP 2, phosphorylated (Tyr542) (SHPTP-2, PTP2C, SH-PTP2, PTP1D, SH-PTP3, Syp, PTPN11)

SHP2 (SH2-domain containing tyrosine phosphatase 2, also known as SHPTP2) is a ~70kD tyrosine phosphatase that plays an important role in cell growth and transformation and mediating integrin, Src and receptor tyrosine kinase (RTK) signaling. SHP2 undergoes tyrosine phosphorylation on several sites including tyrosines 542 and 580 upon binding to platelet-derived growth factor receptor beta (PDGFRb), in response to PDGF stimulation. Tyrosine 542 mediates SHP2 binding to the SH2 domain of Grb2, which in turn couples RTKs to Ras. Phosphorylated tyrosine 542 binds intramolecularly to the SH2 domain in the NH2 terminus of SHP2 and relieves phosphatase inhibition and mediates SHP2 activation of MAPK kinase pathways. \n\nApplications: \nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:1000\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nNIH3T3 cells treated with PDGF\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
