Cat no: S1014-09N

SK3 (SKCa3, KCNN3, K Ca 2.3, Small Conductance Ca2+-activated Potassium Channel 3)

SK3 (KCa3, Kcnn3, SKCa3) forms a voltage-independent potassium channel activated by intracellular calcium. Activation is followed by membrane hyperpolarization. Thought to regulate neuronal excitability by contributing to the slow component of synaptic afterhyperpolarization. The channel is blocked by apamin.\n\nSize: 736 amino acids; 82026D \n\nSubunit: Hetero-oligomer. The complex is composed of 4 channel subunits each of which binds to a calmodulin subunit which regulates the channel activity through calcium-binding (By similarity).\nSubcellular Location: Membrane; Multi-pass membrane protein.\n\nSimilarity: SwissProt: Q9UGI6. Belongs to the potassium channel KCNN family.\n\nEntrez Gene Number: Action potentials in vertebrate neurons are followed by an afterhyperpolarization (AHP) that may persist for several seconds and may have profound consequences for the firing pattern of the neuron. Each component of the AHP is kinetically distinct and is mediated by different calcium-activated potassium channels. The protein encoded by this gene is activated before membrane hyperpolarization and is thought to regulate neuronal excitability by contributing to the slow component of synaptic AHP. The encoded protein is an integral membrane protein that forms a voltage-independent calcium-activated channel with three other calmodulin-binding subunits. This gene contains two CAG repeat regions in the coding sequence. It was thought that expansion of one or both of these repeats could lead to an increased susceptibility to schizophrenia or bipolar disorder, but studies indicate that this is probably not the case. This gene is a member of the KCNN family of potassium channel genes. Two transcript variants encoding two different isoforms have been found for this gene. One of the variants lacks the CAG repeat regions.Action potentials in vertebrate neurons are followed by an afterhyperpolarization (AHP) that may persist for several seconds and may have profound consequences for the firing pattern of the neuron. Other applications not tested.\n\nRecommended Dilution: \nWestern Blot: 1:200 using ECL on rat brain membranes.\nImmunohistochemistry: Rat brain sections.\nFurther dilutions should be made using a carrier protein such as 1-3% BSA or HSA.\nOptimal dilutions to be determined by the researcher.\n\n\nControl Antigen: \nCat#S1014-09N1\n\nControl Antigen Included: 20ug of lyophilized peptide (MW: 2172D)\nControl Antigen Storage before Reconstitution: Lyophilized powder can be stored intact at RT for several weeks. For longer periods, it should be stored at -20 degrees C.\nControl Antigen Reconstitution: 100ul water.\nControl Antigen Storage after Reconstitution: -20C.\nPreadsorption Control: 1ug peptide per 1ug antibody.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
