STAT 1, phosphorylated (Alexa Fluor(R) 488)

Cat no: S7969-10A

STAT 1, phosphorylated (Alexa Fluor(R) 488)

Stat1, while activated in response to a large number of ligands (1), appears to be essential for responsiveness to IFN-alpha and IFN-gamma (2,3). Phosphorylation of Stat1 at Tyr701 induces Stat1 dimerization, nuclear translocation and DNA binding (4). Stat1 has two isoforms, Stat1apha (91kD) and the splice variant Stat1beta (84kD). In most cells, both isoforms are activated by IFN-alpha, but only Stat1alpha is activated by IFN-gamma. Stat1 has been found to be inappropriately activated in many tumors (5). In addition to tyrosine phosphorylation, Stat1 is phosphorylated through a p38 mitogen-activated protein kinase (MAPK)-dependent pathway at Ser727 in response to IFN-alpha and other cellular stresses (6). Serine phosphorylation may be required for the maximal induction of Stat1-mediated gene activation. \n\nApplications: \nSuitable for use in Flow Cytometry. Other applications not tested.\n\nRecommended Dilution:\nFlow Cytometry: 1:10, Add 10ul to 500,000 cells in 90ul PBS, 0.5% BSA.\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C. For long-term storage, aliquot and store at 4 degrees C. Do not freeze. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. Protect from light.\nNote: We do not recommend adding glycerol as a cryopreservative.
