STATs were originally identified as two novel DNA-binding proteins (STAT1 and STAT2) which were found to play essential roles in interferon IFNa- and IFNg-regulated gene expression (2). In unstimulated cells STAT proteins exist largely in the cytoplasm as latent transcription factors. In response to treatment of target cells with cytokines or in some cases growth factors, specific STATs undergo tyrosine phosphorylation, homo- or heterodimerization, nuclear translocation, and DNA binding resulting in the transcriptional activation of distinct target genes (2,3). At least one and oftentimes several STAT proteins are activated in response to all cytokines which utilize cytokine receptor superfamily members. Nevertheless, a striking specificity of STAT activation is seen in response to individual cytokines (3). STAT3 (or APRF- for acute phase response factor) was the first of the second series of STAT proteins to be identified. The 89kD STAT3 protein is expressed in most cell types and is activated by multiple cytokines and growth factors including: IFNa, IL-10, IL-6, IL-11, onM, LIF, CNTF,G-CSF, IL-12, IL-2, GH, EGF, PDGF, and CSF-1 (2). STAT3 binds to DNA as either a homodimer or as a heterodimer with STAT1 (1,2,3,6,7).\n\nApplications: \nSuitable for use in ELISA, Western Blot, Immunoprecipitation and Gel Mobility Shift Assay. Other applications not tested.\n\nRecommended Dilution:\nELISA: 0.1-1ug/ml\nWestern Blot (8): 1-2ug/ml\nImmunoprecipitation (8): 5ug \nGel Mobility Shift Assay (8): 0.05-1.0ug\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nHuman A431 cells (+/- EGF treatment), WI-38 human fibroblasts, NIH 3T3 cells and 293T cells transfected with a mouse STAT3 expression plasmid.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
