Synapsin II

Synapsin IIa and IIb are 74 and 54kD proteins that coat synaptic vesicles (2). Their homolog, synapsin I, may function as an adaptor that tethers the reserved pool of synaptic vesicles to the cytoskeleton meshwork. Depletion of synapsin II expression by antisense oligonucleotides prevents axon elongation in cultured rat hippocampal neurons (3). Depletion of this protein also causes abnormal distribution of intracellular filamentous actin and decreases in the amounts of several synaptic vesicle-associated proteins (3). Genetic studies have shown that mice lacking synapsin I and II are viable but experience seizures with a frequency proportional to the number of mutant alleles (4). Synapsin II knockout mice exhibit decreased post-tetanic potentiation and severe synaptic depression upon repetitive stimulation. This phenotype can be explained by synapsin serving as a regulator in providing fusion-competent synaptic vesicles at the active zone during increased rate of vesicle transport (4). Dephosphorylated synapsin II induces actin polymerization whereas PKA-dependent phosphorylation reduces its ability to interact with actin monomers and filaments (5).
