TAG 72 (CA 72-4, Tumor-associated Glycoprotein 72) (HRP)

Tumor-associated glycoprotein (TAG-72) is an oncofetal antigen with properties of a mucin. Anti-TAG-72 defines the mucin-carried sialylated-Tn epitope. A majority of human adeno-carcinomas including colorectal, pancreatic, gastric, ovarian, endometrial, mammary, and non-small cell lung cancer display some cell populations that are positive for TAG-72 staining. Weak or no reactivity has been observed with most cell types of normal adult tissue excepting the secretory endometrium. About 60% of carcinoma patients express TAG-72 in their sera. It is reportedly useful in distinguishing pulmonary adenocarcinomas from pleural mesotheliomas.\n\nApplications:\nSuitable for use in Western Blot, Flow Cytometry, Immunoprecipitation, Radioimaging and Immunohistochemistry. Other applications not tested. \n\nRecommended Dilution: \nWestern Blot: 1-2ug/ml for 2hrs at RT\nImmunoprecipitation: Use Protein G, 2ug/mg protein lysate\nImmunohistochemistry, Formalin-fixed, paraffin-embedded: 0.2-0.4ug/ml for 30 min at RT. Staining of formalin-fixed tissues REQUIRES boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min.\nOptimal dilution determined by the researcher. \n\nEpitope: Mucin-carried-sialylated-Tn epitope (B72.3); Mucin-carried carbohydrate epitope Gal Beta(1-3) [NeuAcalpha(2-6)]GalNAc (CC49)\n\nPositive Control: Ls174T cells. Colon Carcinoma\n\nCellular Localization: Cell surface and cytoplasmic.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 6 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n
