The Vesicle Associated Membrane Proteins (VAMP) or synaptobrevins are calcium binding proteins specific to eukaryotes. VAMPs, along with synaptosomal associated protein of 25kD (SNAP-25) and syntaxin, form the core complex of soluble NSF attachment protein receptor (SNARE) proteins that interact with the soluble proteins N-ethylmaleimide-sensitive factor (NSF) and alpha-SNAP. These membrane associated proteins play a key role in the regulation of vesicle membrane fusion with the plasma membrane. The Clostridium tetani neurotoxin is a metalloprotease with specificity for VAMP. In Alzheimer?s disease, VAMP levels of all isoforms appear to be significantly lowered. It has been shown that the extreme carboxy-terminus of VAMP-1 is involved in subcellular vesicle targeting. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile glycerol (40-50%), aliquot and store at -20 degrees C. Aliquots are stable for at least 3 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
