Vasodilator Stimulated Phosphoprotein (VASP)

VASP (vasodilator stimulated phosphoprotein) plays an important role in ANF/NO/cGMP Protein kinase and cAMP/cAMP Protein kinase signalling pathways. VASP is expressed in almost all human and animal cell lines; particularly high concentrations are found in thrombocytes, vascular smooth muscle cells and fibroblasts. In cultured cells VASP is associated with focal contacts, cell-cell-contacts, microfilaments and dynamic membrane regions such as the leading edge. In vitro binding data show that VASP binds to profilin, zyxin, vinculin, and the Listeria spp. surface protein ActA. Functional evidence indicates that VASP is a crucial factor involved in the enhancement of actin filament formation. Ser 157 is a site preferred by cAMP-dependent protein kinase (PKA). \n\nApplications:\nSuitable for Western Blot, Immunoprecipitation and Immunocytochemistry. Other applications have not been tested. \n\nRecommended Dilutions: \nImmunocytochemistry: 1ug/ml, formaldehyde fixed cells\nImmunoprecipitation: 2ug/ml\nWestern Blot: 0.1ug/ml\nOptimal dilutions to be determined by researcher. \n\nPositive Control (included):\nHuman platelet protein (500ug), supplied at 5mg/ml in SDS stop solution (100mM NaCl, 73mM TRIS/HCl, pH 6.7, 10mM DTT, 8mM EDTA, 5% glycerol, 2% SDS, 10ug/ml Bromophenol Blue sodium salt. \nUse 5ul (25ug) per lane for Western blotting of tricine gels (13% acrylamide) or 15% Laemmli gels.
