Bad (Bcl2 Antagonist of Cell Death)

Bad is a 22kD protein that activates apoptosis. It exhibits sequence homology to Bcl-2, a protein that prevents apoptosis, within its BH1 and BH2 domains (1). Bad heterodimerizes with Bcl-Xl and Bcl-2 and reverses the apoptosis suppression effect by Bcl-Xl but not Bcl-2. Bad is phosphorylated by Raf-1, a serine-threonine kinase that is targeted to the mitochondrial membranes by Bcl-2 (2). In the presence of survival factor IL-3, Bad is phosphorylated on two serine residues within its 14-3-3 consensus binding sites. Phosphorylated Bad is then sequestered in the cytosol by binding to 14-3-3 and unable to heterodimerize with Bcl-Xl on mitochondrial membranes to promote apoptosis (3).\n\nApplications:\nSuitable for use in Western Blot and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot (ECL): 1ug/ml \nImmunohistochemistry: 1:50\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nRat Brain Tissue Extract\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage, store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Recognizes mouse BAD at ~23kD. Species Crossreactivity: monkey, rat, hamster, bovine, sheep, porcine, canine (weak), rabbit, chicken (weak) and Drosophila. This 23kD band can be specifically inhibited in peptide competition studies.
