Human CD33 is a 67kD sialoadhesin belonging to the Ig superfamily. It is expressed on normal myeloid cells and precursors, as well as myeloid leukemia cells, but is not present on stem cells. Antibody WM-53 is not cytotoxic, and has no detectable effect on hemopoetic cell progenitor assays.
Applications:
Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilutions:
Flow Cytometry: 5ug/ml
Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol:
1. 5x10e5 cultured human HL60 cells were pre-incubated for ~5min with 20ul of 250ug/ml human Ig (to block non specific binding) after which they were incubated 45 minutes on ice with 80ul of C2385-02M at 5ug/ml.
2. Cells were washed twice and incubated with IgG (FITC) goat x mouse , after which they were washed three times, fixed and analyzed by FACS.
3. Cells stained positive with a mean shift of 1.22 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
Storage and Stability:
May be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
