CIITA regulates basal and interferon induced MHC-II transcription through interactions with multiple proteins including transcription factors, chromatin modifying enzymes and components of the basal transcription machinery. CIITA regulates MHC-II expression principally in immune cells, but can also affect their expression in endothelial and fibroblast cells. Mutations in this gene have been associated with bare lymphocyte syndrome type II, which leads to an extreme susceptibility to viral, bacterial and fungal infections. Silencing of CIITA through epigenetic mechanisms also has a major impact on MHC-II expression in tumor cells and can facilitate immune escape. Applications: Suitable for use in ELISA and Immunoprecipitation. Other applications not tested. Recommended Dilution: Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, add sterile 40-50% glycerol, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
