D,L-Citrulline, Conjugated

Citrulline exists primarily in the liver, where it is involved in the urea cycle to detoxify and excrete ammonia. This unusual amino acid is formed in the urea cycle by the addition of carbon dioxide and ammonia to ornithine. It is combined with aspartic acid to form arginosuccinic acid, which later is metabolized into the amino acid arginine. In bacteria, the arginine dihydrolase pathway deaminates arginine to citrulline and splits citrulline into ornithine and carbamoyl phosphate. This pathway is a source of energy for many microorganisms and is used as an identifying test. Citrulline is not a component of any major proteins or enzymes. \n\nApplications:\nSuitable for use in ELISA and Immunohistochemistry. Other applications not tested.\n\nRecommended Dilutions:\nImmunohistochemistry: 1:1000-1:5000.\nOptimal dilutions to be determined by the researcher.\n\nCrossreactivity:\nUsing a conjugate Citrulline-Glutaraldehyde-Protein, antibody specificity was performed with an ELISA test by competition experiments with the following compounds:\nCompound Crossreactivity ratio (a)\nCitrulline-G-BSA 1\nHomo-Citrulline-G-BSA 1:1000\nArginine-G-BSA >1:100,000\nGlutamine-G-BSA >1:100,000\nOrnithine-G-BSA >1:100,000\n\n(a): Citrulline-G-BSA concentration/unconjugated or other conjugated concentration at half displacement\nG = Glutaraldehyde, BSA = Bovine Serum Albumin\n\nStorage and Stability:\nLyophilized powder may be stored at -20 degrees C. Stable for 12 months at -20 degrees C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Reconstituted product is stable for 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
