IRAK 2 (IL-1 Receptor-associated Kinase)

Interleukin-1 receptor-associated kinases (IRAKs) are upstream activators of NFkB in the inflammatory response mediated by IL-1R and Toll-like receptors (TLRs) after exposure to IL-1 or bacterial lipopolysaccharide(LPS). The first human IRAK to be cloned was IRAK11, followed by IRAK22, IRAK-M3 and IRAK44. The IRAKs share sequence homology to the Drosophila protein kinase pelle. Each contains a death domain (DD), which is used for protein-protein interactions with the DDs of other molecules, and putative kinase domains. IRAK2 is catalytically inactive due to the absence of key residues in its putative kinase domain. It is known to associate with the adaptor protein MyD88 in the IL-1R/IRAK/TRAF6/ NFkB signaling pathway, as well as with Akt (protein kinase B) and with Ras in the Ras/IL-1L/p38 MAP kinase signal transduction cascade. IRAK2 and MyD88 are required for IL-1R induced NFkB activation; therefore both may represent therapeutic targets for inhibiting IL-1-induced inflammation.\n\nApplications:\nSuitable for use in Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1-3ug/ml\nOptimal dilutions to be determined by the researcher.\n\nPositive Controls:\nHuman K562, HeLa, HepG2, PC-3, HEK293 and BC-1 cell lysates.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

Additional Info

Recognizes human IRAK2 protein. On Western blots, it identifies the target band at ~65kD. Another band at ~130kD may represent a dimer. In K562 cell lysates, a band at ~80kD has also been observed.
