MSK1 (Mitogen and Stress activated protein Kinase1)

Members of the p90 ribosomal S6 kinase (RSK) family of serine/threonine kinases are substrates of the Ras/extracellular signal-regulated kinase (ERK) pathway, which regulates a variety of cellular processes, including cell proliferation, survival, growth, and motility. RSKs assist in the regulation of transcription factors and coregulators such as CREB, CREB-binding protein (CBP), p300, c-Fos, and estrogen receptor. The RSK family consists of four isoforms and two structurally related proteins, MSK1 and MSK2. Mitogen and stress response kinase-1, or MSK1, is constitutively localized in the nucleus where it interacts with the general transcription activators CBP and p300. These interactions have potential implications in ontogenesis and tumor formation. MSK1 is also activated by p38 and transduces downstream effects of p38. MSK1 may play an important role in TGF-b-mediated responses through p38alpha and Smad signaling pathways. MSK1 and MSK2 are rate limiting for the transcription of COX-2 and IL-1beta genes, suggesting that they may play a role in these processes, and thus, are potential targets for the development of novel anti-inflammatory drugs.\n\nApplications:\nSuitable for use in Western Blot and Immunoprecipitation. Other applications have not been tested.\n\nRecommended Dilution:\nWestern Blot: 1-3ug/ml\nImmunoprecipitation: 0.5ug/IP reaction\nOptimal dilutions to be determined by the researcher.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer. \n

Additional Info

This antibody recognizes the ~90kD MSK1 protein. Bands of unknown origin were observed at ~50kD and ~130kD in Western blots of various cell lysates. This antibody does not recognize the related MSK2 protein. Reactivity has been confirmed with human MSK1 transfected-HEK293, A375, SK-MEL-19, SK-MEL-37, SK-MEL-187, JEG3, NCI-H23, and NCI-H69 cell lysates.
