p130Cas (Cas for Crk-associated substrate, BCAR1, K-associated Substrate, Breast Cancer anti-estrogen Resistance 1 Protein)

The p130Cas (Cas for Crk-associated substrate) is a common cellular target of phosphorylation signal via v-Crk and v-Src oncoproteins. p130Cas has a unique structure that contains a Src homology (SH)-3 domain followed by multiple YXXP motifs and a proline-rich region. p130Cas is implicated in a variety of biological processes including cell adhesion, cell migration, growth factor stimulation, cytokine receptor engagement and bacterial infection. Physiological functions of p130Cas include cardiovascular development, actin filament assembly and Srcinduced cell transformation. \n\nApplications:\nSuitable for use in Immunoprecipitation, Western Blot and Immunhistochemistry. Other applications not tested. \n\nRecommended Dilutions: \nImmunoprecipitation (Denatured verified): 2ug/mg protein lysate. (Use Protein G).\nWestern Blot:1-2ug/ml for 2hrs at RT \nImmunohistochemistry (Formalin/paraffin): 1-2ug/ml for 30 min at RT. Note: Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 min. \nOptimal dilution determined by the researcher. \n\nMW of Antigen: 130kD \n\nPositive Control:\nMCF-7 cells, HUVEC cells. Tonsil or breast carcinoma. \n\nCellular Localization: Cytoplasmic\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at-20 degrees C. Aliquots are stable for at least 12 months at-20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
