p130Cas (Cas for Crk-associated substrate, BCAR1, K-associated Substrate, Breast Cancer anti-estrogen Resistance 1 Protein)

A 130kD protein, designated p130, represents one of several known substrates for v-Crk encoded p47. p130 (designated Cas for Crk associated substrate) exhibits a high level of tyrosine phosphorylation and is tightly associated with v-Crk, suggesting a role in v-Crk mediated cell signaling. The molecular cloning of Cas p130 has shown to represent a novel SH3 containing signaling molecule with a cluster of multiple putative SH2-binding motifs for v-Crk. By immunoprecipitation analysis, Cas p130 has been shown to be highly phosphorylated at tyrosine residues subsequent to either v-Src p60 or v-Crk- mediated transformation and to form stable complexes with both of these transforming proteins. Cas p130 behaves as an extremely potent substrate for protein tyrosine kinases and has been reported to relocate from the cytoplasm to cell membrane upon tyrosine phosphorylation. One proposed model is that the SH2 domain of v-Crk function to activate c-Src kinase, which in turn phosphorylates Cas p130.\n\nApplications:\nSuitable for use in Western Blot. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1-2ug/ml for 2h at RT\nOptimal dilutions to be determined by the researcher.\n\nPositive Control:\nSkeletal muscle, T47D cells\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
