Phosphoserine

Phosphorylation and dephosphorylation of cellular proteins are central steps in transducing extracellular signals to the cell nucleus. Phosphorylated epitopes may serve as docking sites for the assembly of protein complexes or may alter the 3-dimensional protein structure thus modulating enzymatic activity or the ability to undergo protein-protein-interactions. Modification of proteins on serine residues is mediated by serine/threonine kinases.\n\nApplications:\nSuitable for use in ELISA, Immunoprecipitation and Western Blot. Other applications have not been tested.\n\nRecommended Dilution:\nELISA: 0.05ug/ml\nImmunoprecipitation: 1-10ug per 10e6 pervanadate-treated A431 cells\nWestern Blot: 1ug/ml for AP/BCIP/NBT (MTT) detection; 0.1ug/ml for HRP/ECL detection. BSA/Tween 20 based blocking buffer. Do NOT use milk/casein for blocking and dilution.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control Included:\nP4076-22A: Phosphoserine/Phosphothreonine, Positive Control\nSupplied as a liquid in PBS, 10mM NaF, PEG, sucrose, 0.09% sodium azde. Add 75ul SDS-PAGE sample buffer and incubate at 90 degrees C for 5 minutes.\nWestern Blot: 20ul/lane for HRPO/ECL detection. 5ul/lane for HRPO/DAB detection.\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
