BNIP3 (Bcl2/Adenovirus E1B 19kD Protein-Interacting Protein 3, 19kD Interacting Protein 3, NIP3)

BNIP3, formerly NIP3 (nineteenkD interacting protein-3), is a pro-apoptotic, mitochondrial protein. It represents a subfamily of Bcl 2 related proteins, which functions without a typical BH3 domain to regulate apoptosis from both mitochondrial and non-mitochondrial sites by selective Bcl 2/Bcl XL interactions. It interacts with the E1B 19kD protein, which is responsible for the protection of virally-induced cell death, and the E1B 19kD-like sequence of Bcl2, also an apoptotic protector. BNIP3 also plays a role in autophagy. Hypoxia-induced autophagy requires expression of this protein. Beclin, an autophagy protein, is kept in check by Bcl2 (normoxic). When Bcl2 lets go of Beclin it then binds BNIP3/L3 (hypoxic), causing autophagy. \n\nAlternate Names: anti-BCL2/adenovirus E1B 19kD protein-interacting protein 3 antibody \n\nApplications: \nSuitable for use in ELISA, Western Blotting. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 1:500-1:1,000. Hypoxic samples at ~30 and 60kD, representing the BNIP3 protein. There are some non-specific bands present, as well. It is recommended that normoxic samples are run next to the hypoxic samples, as a negative control. \nOptimal dilutions to be determined by the researcher.\n\nCellular Localization: Mitochondrion. Co-expression with the EIB 19kD protein results in a shift in NIP3 localization to the nuclear envelope. \n\nPositive Controls: \nHypoxic Hep3b lysates \n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
