STAT3, phosphorylated (Tyr705) (Signal Transducer and Activator of Transcription 3, Acute-phase Response Factor, APRF) (Biotin)

The STAT proteins serve as both cytoplasmic signal transducers and nuclear activators of transcription. STATs are mediators invovled in cytokine signaling. In response to a specific cytokine signal, STAT proteins are phosphorylated on conserved tyrosine residues. Phosphorylated STAT proteins dimerize via their SH2 domains and move to the nucleus. The STAT dimers bind to specific DNA elements resulting in transcriptional regulation of downstream target genes. \nSTAT3 is activated by tyrosine phosphorylation at residue Tyr705 in cells treated with Interleukin 6 or EGF. Activated STAT3 can bind to DNA either as a homodimer or as a heterodimer with STAT1.\n\nApplications: \nSuitable for use in Western Blot, Immunoprecipitation, Immunocytochemistry, Immunohistochemistry and ELISA. Other applications not tested.\n\nRecommended Dilutions:\nWestern Blot: 0.5ug/ml for HRPO/ECL detection. Recommended blocking buffer: Casein/Tween 20 based blocking and blot incubation buffer. \nImmunoprecipitation: 1-10ug per 10e6 pervanadate treated HepG2 cells.\nImmunocytochemistry: 1-10ug/ml\nELISA: 0.05ug/ml\nOptimal dilutions to be determined by the researcher. \n\nPositive Control Included:\nH2030-03H1: HepG2 Cell Lysate, Vanadate Treated\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for at least 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
