Trimethylation of histone H3 on Lys9 (H3K9me3) is one of the most highly studied epigenetic marks. H3K9me3 functions in the repression of euchromatic genes, and in epigenetic control of heterochromatin assembly, most likely by acting as a recognition motif for the binding of chromatin-associated proteins, such as Swi6 or HP1a. The enzymes responsible for H3K9 trimethylation are SUV39H1 and SUV39H2. Suv39H1 is a nuclear protein that acts as a H3K9 specific methyltransferase (HMT). In addition to its role in gene silencing, Suv39H1 is also thought to be involved in heterochromatin organization, chromosome segregation, and mitotic progression, and the epigenetic regulation of telomere length in mammalian cells.\n\nApplications:\nSuitable for use in Western Blot and Immunoprecipitation. Other applications not tested.\n\nRecommended Dilution:\nWestern Blot: 0.1ug/ml \nImmunoprecipitation: 10ug\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nU2OS lysate\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and store at -20 degrees C. Aliquots are stable for at least 12 months at -20 degrees C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
