Vascular Endothelial Growth Factor, VEGF (VG1)

Angiogenesis, the formation of new vessels, is a highly regulated process. There is strong evidence that Vascular Endothelial Growth Factor (VEGF), also known as vascular permeability factor (VPF), is an important and major regulator both of physiological and pathological angiogenesis. VEGF is a dimeric glycoprotein (45kD) that is structurally related to PDGF. VEGF is synthesized by both tumor and normal cells and, unlike many other growth factors that are broadly active, acts specifically on endothelial cells. The VEGF gene contains 8 exons. Analysis of VEGF cDNAs predicts three alternately spliced VEGF protein isoforms: VEGF121 (lacking exons 6 and 7), VEGF165 (lacking exon 6) and VEGF189. VEGF binds to the endothelial tyrosine kinase receptors FLK1 and FLT1. In addition, neuropilin-1 binds specifically to the VEGF165 isoform. VEGF stimulates and induces migration and proliferation of endothelial cells, enhances vascular permeability, and thrombogenicity. Tumor angiogenesis is essential for tumor growth and metastasis. Angiogenesis is linked to increased metastasis formation and decreased overall survival with a variety of malignant tumors. It has been reported that VEGF is a useful marker of tumor angiogenesis. VEGF expression correlates with vascularity, metastasis and proliferation of tumors. Thus, high expression of VEGF in tumor tissue may be a indication of poor prognosis.\n\nApplications:\nSuitable for use in Immunohistochemistry. Other applications have not been tested.\n\nRecommended Dilution:\nImmunohistochemistry: Frozen and formalin-fixed, paraffin-embedded tissue sections. Epitope retrieval is required using citrate buffer, pH 6.0.\nOptimal dilutions to be determined by the researcher.\n\nPositive Control: \nColon cancer\n\nStorage and Stability:\nMay be stored at 4 degrees C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20 degrees C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
